Twifo culture follows the wider Akan pattern: a matrilineal system of inheritance, reverence for ancestors (Nananom), and a chieftaincy institution built around stools held by an Omanhene, supported by divisional chiefs, queen mothers, and councils of elders. Twi is the everyday language, and Twifo speakers regard their dialect as among the purest spoken forms of Twi.

The area's central cultural celebration is the Twifo Guakro Odwira Afahye, an annual festival of purification and renewal. Festivities typically include a visit to the ancestral shrine to welcome participants safely, an Akwaaba (welcome) night of bonfires, drumming, and dancing, and a grand durbar at which chiefs and queen mothers are paid homage and the community's history and resilience are publicly recounted. The festival serves both as a homecoming for natives living away and as a living classroom that passes on Twifo identity, customs, and history to younger generations.
